Introduction
This product is a nitlurea antibiotic, mainly used in pancreatic islet cell carcinoma (beta cell or non-beta cell carcinoma). It has a certain effect on carcinoid tumor, Hodgkin's disease, colon cancer and liver cancer. It was an effective DNA methylation reagent for HL60, K562 and C1498 cells with IC50 values of 11.7, 904 and 1024μg/mL, respectively. The laboratory is often used to replicate diabetes models.
This product is a nitrourea antibiotic produced by Stre. Achromogenes Uar.128. It differs from the fat-soluble nitrite in that it is a methyl group at the ethyl chloride and an amino sugar at the other end of the molecule. STZ can decompose active positive methyl carbon ions and form interstrand cross links with DNA to alkalize DNA. However, the alkylation of STZ is weaker than that of other nitlurea drugs, while the alkylation of its metabolite methylnitlurea is 3~4 times stronger than that of STZ. STZ can form isocyanates in vivo. Thus, it binds with nucleic acid protein, inhibits the activity of DNA polymerase, and makes the damaged DNA difficult to repair. In the process of anti-tumor research, it was found that STZ could increase blood glucose in mice, and cause diabetes in dogs and monkeys, and it was permanent. The diabetic effect of STZ has different species and does not cause diabetes in guinea pigs and humans. The mechanism of diabetes is mainly due to the decrease of nicotinamide adenine (DNA) content in islet cells. The glucose base in STZ molecule can make STZ enter islet β cells, cause morphological changes in the nucleus of β cells, and make their chromosomes agglutinate, elongate and concentrate.

Instruction
A diabetes model was used as an example to illustrate the use of STZ. 1. Preparation of citric acid buffer liquid A: 2.1 g of citric acid (MW:210.14) was weighed and added into 100 mL of double steaming water to prepare liquid A; Liquid B: sodium citrate (MW:294.10) 2.94g was added to 100 mL of double steaming water to form liquid B. Mix A and B at a certain ratio (1:1.32 or 1:1), measure the pH value, adjust the pH in the range of 4.2-4.5, that is, the citric acid buffer needed to prepare STZ. 2. Before preparing STZ injection, place STZ in a dry and sterilized bottle, wrap it with tin foil, put it in an ice bath with citric acid buffer, and bring it to the animal room for later using. 3. The rats prepared for injection were weighed after overnight fasting. The rats were grouped so that STZ could be dissolved according to the group. 1% STZ injection was prepared with citrate buffer according to fasting body weight. If the subsequent injection operation is not skilled, do not dissolve the STZ at one time. Note: STZ is easy to deactivate, STZ is still required to be dry and avoid light after quick weighing, and dry aluminum foil (or tin foil) paper is recommended. 4. Intraperitoneal injection or tail vein injection. If the injection operation technique is not skilled, the two groups should be injected alternately and the injection should be completed within 30 minutes. Note: Most injections require a quick injection.
Note
STZ is unstable under moisture. If it needs to be weighed several times, it should be operated and stored in strict accordance with the principle of avoiding moisture. The operating environment, containers, and packing tools must be kept dry.
Long exposure should be avoided to prevent moisture. It expires after 30 minutes of damp exposure. This is for the same reason that modeling requires rapid injection because the aqueous solution is unstable.
The injection was prepared only before injection because the aqueous solution of STZ was extremely unstable.
