Product Description/Introduction
RNAsolidTM RNA Stabilization Solution for Tissue is commonly used to rapidly inhibit RNase and stabilize RNA in tissue samples at room temperature when collecting animal tissues. This product can be widely used for the preservation of fresh animal tissue samples such as liver, brain, lung, spleen, kidney, heart, muscle, etc. as well as certain plant samples. It is suitable for cases where RNA extraction (including miRNA and other small RNAs) is not possible immediately after sample collection, and it is especially suitable for the stabilization and preservation of RNA in samples collected in the field, animal tissue samples collected in large batches and samples that cannot be frozen immediately. The effect of this product is comparable to that of liquid nitrogen freezing. This product can quickly penetrate into the tissue, completely inhibit endogenous RNase activity, avoid RNA degradation, maintain the integrity of the RNA in the sample, and, like liquid nitrogen, preserve the RNA in the sample for a long time. This product is safe and convenient, without any toxicity and irritation, and can be used at room temperature which ensures the safety and convenience of use. Compared with the use of liquid nitrogen, it can effectively avoid frostbite, centrifuge tube bursting and other safety hazards caused by the use of ultra-low-temperature liquid nitrogen, and it is very convenient to carry and transport. The storage time of the samples in this product is related to the storage temperature, e.g., 1 to 2 days at 37°C, one week at room temperature 25°C, one month at 4°C, and long-term storage at -20°C or lower.

Assay Protocol / Procedures
1. Sample handling
a) Animal tissue: After cutting the tissue into appropriate sizes (it is recommended that the length, width and height of the tissue block be no larger than 0.5 cm), the tissue is then quickly and completely immersed in 5-10 times the volume of RNAsolidTM RNA Stabilization Solution for Tissue. (Note: Tissues that have been immersed in RNAsolidTM Solution can be removed from the solution after a period of equilibration, cut into smaller pieces of tissue, and placed back into RNAsolidTM Solution; some of the more weakly liquid-permeable tissues, such as bone, are not suitable for RNA protection with this product.)
b) Plant tissue: This product can directly penetrate soft plant tissues (e.g. young leaves, young stems) and achieve ideal RNA protection. Cut the tissues into appropriate sizes (recommended length, width and height not more than 0.5 cm) and then quickly immerse them in 5-10 times the volume of RNAsolidTM RNA Stabilization Solution for Tissue. (Note: Some plant tissues have inherent barriers, such as the wax layer on the surface of leaves, that may impede penetration of RNAsolidTM Solution, and for such tissues it is often necessary to break down these barrier layers to allow penetration of the solution.)
2. Sample preservation
Samples can be stored in RNAsolidTM RNA Stabilization Solution for Tissue at 37°C for 1-2 days,after 2 days some of the RNA in the tissue will begin to degrade. After 1 week of stable storage at room temperature (25), no significant RNA degradation occurs. Most of the samples can be stored in RNAsolidTM RNA Stabilization Solution for Tissue at 4 for 1 month without significant RNA degradation. If long-term storage is required, the samples in RNAsolidTM RNA Stabilization Solution for Tissue should be soaked in the solution overnight at 4, and then the samples should be directly put into -20 or -80, which can be used for long-term and stable storage for 3~5 years (in order to accelerate the thawing at a later stage, it is also possible to suck out the RNAsolidTM RNA Stabilization Solution for Tissue and discard it prior to putting the samples into -20 or -80 for storage).
3. RNA isolationRemove the tissue block from the RNAsolidTM RNA Stabilization Solution for Tissue with clean forceps, and then remove the surface liquid with clean absorbent paper. Immediately afterward, homogenize in a suitable lysis solution or grind in liquid nitrogen, and then proceed to RNA extraction using Trizol or centrifugal column method (G3013 recommended).
Note
1. The tissue samples should be quickly and completely immersed in RNAsolidTM RNA Stabilization Solution for Tissue after proper size processing.
2. Using RNAsolidTM Solution for samples frozen at -20°C or -80°C, repeated freezing and thawing 3-5 times will not significantly affect the quality of RNA. In general, it is recommended that samples preserved in this product be transported or stored at room temperature or 4°C for short-term storage and -20°C or -80°C for long-term storage. For long-term storage at -20 or -80, the samples must be stored at 4 overnight or longer before transferring to a lower temperature for storage. It is prohibited to store the products in liquid nitrogen directly, the reagent tubes are not resistant to low temperature storage in liquid nitrogen, and there is a risk of blowing up.
3. This product is suitable for the stabilization and preservation of RNA in fresh animal tissue. Samples frozen and then preserved in RNAsolidTM will be much less effective than fresh samples preserved directly in RNAsolidTM. This product is more suitable for fresh samples, although it also has some stabilizing effect on frozen samples.
4. The tissue samples stored by this product can be extracted by conventional RNA extraction methods, such as Trizol and centrifugal columns, etc. When used, the samples can be taken out of the RNAsolidTM solution directly as conventional fresh or frozen samples, and a small amount of residual RNAsolidTM solution will not affect the results of the subsequent extractions.
5. After preservation with this product, the sample may become slightly hard, which is normal and will not affect the homogenization and lysis of the tissue during subsequent RNA extraction.
6. -20 or -80 storage, the sample thawing RNAsolidTM solution may have crystals precipitation, is a normal phenomenon, you can wait until the crystals are completely dissolved to use, will not affect the subsequent RNA extraction effect.
7. When used for the preservation of plant samples, surface waxes should be removed to ensure that the RNAsolidTM solution penetrates sufficiently into the tissue.
