Introduction


EDTA is widely used in various fields of biology, such as electrophoresis buffer system, metal dependent proteolytic enzyme or nuclease reaction, cell digestion or sample depolymerization process, etc. EDTA solution has been used in a study to help elucidate the role of antigen repair in immunostaining of ethanol fixed smears. EDTA solution was also used in the study to evaluate the uptake and release of biotin labeled fluorescein (b-fitc). EDTA is often used as an anticoagulant in the form of sodium or potassium salts. The mechanism of action is that it can chelate calcium ions in blood to prevent coagulation. Ethylenediaminetetraacetic acid is a water-soluble divalent ion chelator, which can form complexes with metal ions such as manganese (II), copper (II), iron (III) and cobalt (II) to inhibit metal dependent enzymatic reactions.

1. EDTA and Mg2+, Ca2+, Mn2+, Fe2+ and other divalent metal ions, commonly used to do nuclease, protease inhibitors, in the extraction of mouse liver tissue protein experiments, EDTA can be used for the preparation of Triton lysis buffer, the concentration of EDTA is 1 mM, take 2.92 mg EDTA dissolved in 10 mL water.

2. In tryptic digestion experiments, EDTA can be used to prepare tryptic digests with a solution formulation of 0.15% (wt/vol) trypsin, 2.4 mM EDTA, and 0.0005% (wt/vol) phenol red, dissolved in PBS.
